Well rounded in 6e

400 points is high for a non-super campaign. That said, it depends on the power level you want to run at.

 

At 8-9 DC's and 3-4 SPD, you can simulate the manga/anime pretty well at 250-300 points. Every PC will be weaker than Luffy (thank God) but they'll be as good or better than the sidekicks. If you want everyone to be at Luffy's level, then go ahead and start at 400. The key thing to remember is that every PC should be able to be the star on a rotating basis, not one star with the featured sidekick of the week.

What is well rounded is going to be dependent on the campaign.  What is well rounded in a high-powered superhero campaign is going to be overwhelming in a   low powered street level superhero campaign.   When you start comparing superhero campaigns to heroic campaigns it gets even worse.  

 

The other thing to factor in is how optimized the characters are.  There are a lot of ways to write up effective characters without having to spend huge amount of points.  A lot of player forget about complementary skills .   Sometimes the best way to be the best at something is not just to throw more points at it.  Having the right complementary skills not only gives you those skills, they can in the right circumstance boost up your other skills.  Background skills are particularly good at this and with skill enhancers can be dirt cheap.

Remember that the OP wants to simulate a popular Shonen anime. Low powered supers are the floor here. Most of the characters can be well rounded versions of themselves at 250-300 points but if they all want to be close to the main characters then they will need more. He could start them at 200 and then give experience at a higher rate than is normal in Hero also.

I had a GM once who, after our characters were fully built, gave us an extra 10 free points that had to be spent on "useless" KS's and similar background Skills.  You could tweak the amounts. 

 

It's easier to give than take away, and it's easier to say "You get 175 points plus 10 points worth of free background skills" than it is to say "You get 185 points but you must spend 10 of them on background skills".

Plus, at least the first time, if you say 185 with 10 must be in background skills, they'll put exactly 10 in the background skills.  If you say 175, hey, who knows, some of em might put some points into them during the build.

 

Second time around, of course, the players might expect it, so you might not be able to surprise them...but hey, then maybe you change the deal a bit....

Aww, let's give everyone one 3 point background skill at 75 points. Add a +1 to that skill at 125 points, taking that +1 & adding another skill instead at 150 points; 200 would be 75+75+50 a background skill & a +1 background skill, 225 would be three background skills, no pluses. If your a martial artist, speedster, warrior or rogue add another set for DEX skills rarely used in combat like Acrobatics & Breakfall. Don't even add these to your points, these are rarely if ever used in combat, even the DEX ones, consider them skills you gain from adventuring you shouldn't be penalized for.
