    Not discussing wich is the best, I'm here just to tell I played a campaign in 6th century Europe, from Byzantium to the Vandal Kingdom to the Persian frontier in Armenia, with the Blue and Green hippodrome factions fighting in the streets, Huns still menacing, conspiracies, spies, and a gold mine.     To me the Brain Trust are the epitome of a comic-book supervillain team, as they're an assemblage of diverse concepts which could coexist in no other genre. Another nice thing about them is that they can be used different ways, committing robberies or as hirelings of another villain group to gain funds, or following one of the Overbrain's more grandiose schemes.
     
    I haven't used them extensively, and not as more than hirelings. But I would suggest that you raise Mr. Zombie's Strength into the 50-60 range. He's a little too light weight for his role as "team brick" compared to standard PCs. Maybe eliminate his Teeth and Nails Killing Attacks if you don't want his increased STR to boost them. I'd also recommend buying up his Running and/or Leaping to more than base level, so he can actually close with the opponents he's supposed to fight.

    You must decide if you want a group of PCs with good relationships between them, or not. I usually give my players their PCs built with a group idea, like a mad doctor and his followers (an assistant, a bodyguard, and a fixer), so I would think of monsters in a pack. Or not, then I would give them a medieval version of the Universal Monsters and let them fight for the same goal. Or may be you want Kaijus.

    Well, this idea might have been used before. What if the PCs are not the good guys but some baddies?
    First problem, for me, is how much of evil are they allowed to be without spoiling the fun. Some things are not fun in a game, I won't like the PCs raping peasants and beating baby seals.
    One solution is "comedic evil", but I want a serious approach.
    Another ideas are "evil can turn against itself, sometimes" or "you see, the minions of the Dark Lord are not so different from people working for unethical corporations and bosses they hate just beacause the money".
    So, inspired by the Turakian Age, I have more or less this outline for a long adventure, and for a group of characters.
    The forces of the Dark Lord, Kal-Turak, conquered a city. But they are a mess of orcs, goblins, humans, ogres, a few trolls... Each group with their captains and chieftains. Kal-Turak hopes one leader will unite them, killing the weakest, but one year passed and there are four factions yet. The PCs a legates from the Dragon Lady, serving the Dark Lord, and their mission is to help the best leader of this factions to become the Overlord.
    One of the PCs is conscripted to serve Kal-Turak, and the rest could suspect of him. He battled against the evil forces, but his hometown was conquered and the warriors were offered to choose death or service. Now he is one of the many lovers of the Lady, and has something to gain serving evil.
    Another may be a wizard who considers himself the best candidate to Overlord. Another can be a big ogre who simply follows orders.
    The strongest contender in the city, based on Zarlang Thul, is very suspicious of the PCs and will conspire to have them killed by unknown assassins and blame the opposition.
    This adventure is what comes to my mind by now, it can work with baddies, not will work well if the PCs were goodies.

    If you want to take a cue from the original movie, the winner is not the person who crosses the finish line first, but who has the largest point total when they cross the finish line.
     
    And how can they get points? 1 for each person they kill. 10 for each police car destroyed. A variable number for each crime they assist on the way.

    Because there are multiple courses.  Each driver will have a unique course, concealed from him until he picks up his car for the race.  The mysterious organizers of the event spend weeks determining alternate routes of equal length and approximately the same number of turns, traffic, and other difficulties, and they ensure that there is plenty of path-crossing, periodic points of paralleling (Wow!  I could have written for Marvel! ) and things like that, just to ensure that driver rivalries have an opportunity to express themselves, and of course, for the mayhem two rival drivers may involve themselves in.
     
    More than "X" amount of variation from the predetermined course results in disqualifications, which, of course, can have its own mob-related consequences.  Exceptions have to be made for sections that are genuinely impassable (perhaps some construction has ripped open a road that, three months ago, was perfectly fine) simply because failure to finish the race for any reason but mechanical failure, crashing out, or getting apprehended also have mob-related consequences.
     
    Courses are put into envelopes, selected randomly, and distributed by courier in the wee hours of the morning of the race, along with times and the starting and finish points of that particular course.
     
    Not only does this keep the actual date of the race a secret until the day it happens, it ensures that there is too much action in too many places for police-- car or helicopter-- to realistically stop all of it.  This has the fortunate side effect of making the police into part of the traffic hazard, as cars will stop for the sirens, and road blocks will be thrown up based purely on guesswork and hope.  Roadblocks will be poorly manned and likely even highly-passable, as the police will be spread too thin to reinforce one another.  As to the choppers-- well, there are only so many choppers to being with, the participants are required to keep their cars painted as typical civilian vehicles, and the tunnels, parking garages, and canyons between the skyscrapers provide great places to shake things up, making it difficult to track just who went where.
